INTENDED USE
For the quantification of Procalcitonin (PCT) in human serum, plasma or whole blood. They are for professional use only.
Used for auxiliary diagnosis of heart failure in clinical.
PRINCIPLE
The PCT Quantitative Test Kit matched with Medomics Fluorescent Immunoassay Analyzer is a fluorescence immunoassay for quantitative determination of PCT (procalcitonin) in human whole blood, plasma, and serum specimen.
Procalcitonin (PCT) is a small protein, which comprises 116 amino acid residues with a molecular weight of approximately 12.7 kDa.
PCT is expressed by neuroendocrine cells (including C cells of thyroid, lung and pancreas tissues) and decomposed into (immature) calcitonin, carboxy-terminal peptide and amino-terminal peptide by enzyme digestion. The blood of healthy people only contains a small amount of PCT. PCT will increase significantly after bacterial infection.
Procalcitonin (PCT) is a specific indicator related to bacterial inflammatory response, and also an important indicator for rapid identification of SIRS and sepsis caused by bacterial infection, especially severe sepsis and septic shock. It is of great significance for identification of inflammatory response caused by infection and non-infection (such as autoimmune inflammation, acute
rejection of transplantation). At the same time, PCT detection plays an important role in guiding the rational use of clinical antibacterial drugs, efficacy evaluation and shortening the use time of antibacterial drugs.
MAIN COMPONENTS
The kit contains the corresponding number of test cassettes for each package specification: the test cassette consists of a test strip shell and a test strip, which consists of a binding pad (containing fluorescent microsphere-labelled anti-calcitoninogen monoclonal antibody I and chicken IgY), a nitrocellulose membrane (with the quality control line coated with goat anti-chicken IgY antibody, and the detection line coated with anti-calcitoninogen monoclonal antibody II), sample pads, blotting paper, and a PVC plate.
Dilution solution: composed of 0.01 mol/L phosphate, 0.1% preservative, pH=7.4.

SAMPLE REQUIREMENTS
The Sample types with test cassette include human serum, plasma or whole blood samples Whole blood collection: Blood is collected using sodium heparin, EDTA-K2 or sodium citrate anticoagulation tubes, and the collected blood sample is added and shaken well and set aside.
Samples should not be frozen and should be tested as soon as possible after collection. If the test cannot be completed within 12 hours, it should be stored at 2°C to 8°C for no more than 2 days.
Serum or Plasma collection: Serum and plasma should be separated as soon as possible after blood collection to avoid haemolysis. The serum and plasma should be tested as soon as possible after separation, and if the test cannot be completed within 12 hours, stored at 2-8°C for no more than 7 days. Long-term storage should be frozen at -20°C for no more than 6 months. Samples should not be frozen and thawed more than 2 times.
Samples must be returned to room temperature prior to testing. Frozen samples must be completely thawed, mixed and brought back to room temperature before use, and must not be repeatedly frozen and thawed.
